Rats are various medium-sized, long-tailed rodents. The rat is suitable as a model for physiology, pathology, pharmacology, toxicology, and transplantation experiments, due to its body size, which is approximately ten times larger than that of the mouse, offering unique advantages in surgical procedures and clinical sampling. Rat primary cells are cells isolated directly from specific rat tissue using enzymatic or mechanical methods. They closely mimic the physiological state of cells in vivo and are expected to maintain their in vivo functions under optimal conditions for a short period of time.
Rat primary cells are established as an important tool for in vitro cell-based assays or for generating in vivo models. Meanwhile, primary cells can become cell lines through spontaneous mutation or deliberate modification. However, the primary cells present several advantages over cell lines which can be passaged in vitro indefinitely, including a better representation of the cellular heterogeneity of tissues, a more faithful transcriptomic and proteomic profile and more realistic functional responses. Usually, tissue masses are disaggregated using chemical, mechanical or enzymatic methods to break up tissues and obtain viable cells. The cells are then induced to grow in culture vessels based on cell characteristics.
