The mouse has become the preferred mammalian model. Among the many reasons for this privileged position of mice is their genetic proximity to humans, the possibilities of genetically manipulating their genomes and the availability of many tools, mutants and inbred strains. Mouse primary cells are cells isolated directly from specific mouse tissue using enzymatic or mechanical methods. They closely mimic the physiological state of cells in vivo and are expected to maintain their in vivo functions under optimal conditions for a short period of time.
Mouse primary cells are established as an important tool for in vitro cell-based assays or for generating in vivo models. Primary cells can become cell lines through spontaneous mutation or deliberate modification. The primary cells present several advantages over cell lines which can be passaged in vitro indefinitely, including a better representation of the cellular heterogeneity of tissues, a more faithful transcriptomic and proteomic profile and more realistic functional responses.
Primary cell culture refers to the culturing of cells directly isolated from tissue or organs. Tissue masses are disaggregated using chemical, mechanical or enzymatic methods to break up tissues and obtain viable cells. The cells are then induced to grow in culture vessels based on cell characteristics.
