Primary cells refer to cells that are isolated or harvested directly from living tissue or organs. With the exception of stem cell and progenitor populations, the majority of primary cells are terminally differentiated cells with a highly specific type and high level of function. The cells can be isolated and cultured in vitro and undergo limited cell divisions before entering senescence.
Primary cells are established as an important tool for in vitro cell-based assays or for generating in vivo models. Meanwhile, primary cells can become cell lines through spontaneous mutation or deliberate modification. However, the primary cells present several advantages over cell lines which can be passaged in vitro indefinitely, including a better representation of the cellular heterogeneity of tissues, a more faithful transcriptomic and proteomic profile and more realistic functional responses.
Usually, tissue masses are disaggregated using chemical, mechanical or enzymatic methods to break up tissues and obtain viable cells. The cells are then induced to grow in culture vessels based on cell characteristics.
