Antibodies play an integral role in the immune defense against various pathogens. Due to the specificity and high affinity for their antigens, antibodies have been a valuable and indispensable tool in research. Today, various antibody products are marketed for research. They are potent tools for biochemical and biomedical research.

Antibody molecules are secreted as globular proteins. An antibody that can bind to a specific protein or a biomolecule of interest is called a primary antibody. Primary antibodies are produced as monoclonal or polyclonal antibodies usually using animal species such as the mouse, rabbit, rat and goat as hosts. Monoclonal antibodies specifically detect a particular epitope on the antigen and therefore are less likely than polyclonal antibodies to cross-react with other proteins.

Fig.1 Schematic drawing of natural rabbit antibodies in IgG format. The ~150-kDa rabbit IgG molecule contains two identical

κ (white) or λ (light gray) light chains paired with two identical heavy chains (dark gray). Notably, rabbits have two κ light chains,

K1 and K2. The more frequent κ light chain, K1, contains an additional disulfide bridge (orange) that links VL and CL. Rabbits of

the commonly used New Zealand White strain have ~90% IgG-κ (K1), ~10% IgG-κ (K2) and <1% IgG-λ antibodies.

Rabbit monoclonal antibodies (mAbs) have two major advantages compared with other mAbs, such as mouse and rat mAbs, in terms of high affinity and specificity. The KD of Rabbit mAbs is typically at picomolar level, while most mouse mAbs at nanomolar or sub-nanomolar level. The highly diverse rabbit immune system enables rabbit mAbs to distinguish very similar proteins or sequences. Furthermore, the rabbit immune system is able to recognize a wider range of epitopes, novel epitopes and small epitopes, even antigens that are not immunogenic in mice. So, rabbit monoclonal antibodies can be a good choice for your projects, such as anti-idiotype antibody development, immunohistochemistry (IHC), flow cytometry (FC) and immunofluorescence (IF).
