Anti-idiotype antibody (Anti-ID Abs) is an antibody against the antigen binding site (complementarity determining region, CDR region) of another antibody. Because most monoclonal antibodies developed today are human or humanized, the most likely immunogenic epitopes for the induction of ADA lie within the hypervariable complementarity determining regions (CDR) that provide the majority of the binding contacts.
We have extensive experiences in developing highly sensitive and highly specific anti-idiotypic antibodies (Anti-ID Abs) to support your antibody-drug development by suing our antibody phage display technologies. Our data shows that recombinant monoclonal antibodies from our antibody phage display libraries are comparable to traditional monoclonal antibodies in their functionality. However, since they are generated using fully in vitro processes they offer greater flexibility during their production and more opportunities for optimization, such as affinity maturation and conversion to different formats.
For anti-idiotypic antibody (Anti-ID antibody) generation using phage display library technology, we have two strategies: 1) panning of the antibody is carried out on the small molecule in the presence of isotype sub-class matched antibodies as blockers, to avoid enrichment of specificities that bind to other regions of the antibody small molecule and to ensure idiotope specificity; 2) panning of high-specifity anti-idiotypic antibody by subtracting the concentration of idiotypic antibody (as a bait) and purified CDRs domain. Also, we will utilize an appropriate immunization and purification screening strategies to develop a successful anti-ID polyclonal and monoclonal antibody for our clients.
