Based on the finding that cells producing antibodies to certain antigens such as viruses, bacterium and parasites circulate in the peripheral blood of pathogen infected individuals, Alpha Lifetech Inc. develops a unique platform to develop fully human monoclonal antibodies from lymphoblastoid cell lines. Lymphoblastoid cell line (LCLs) derives from the fusion cells (here, often refer to B cells) infected by Epstein-Barr virus (EBV). PBMCs (Peripheral Blood Mononuclear Cells), containing the memory B cells, are extracted from whole blood using a density gradient method and activated with different antigens e.g., viral, parasite bacterial antigens. This technology offers a biggest advantage that one can immunize the lymphocytes with those antigens, which are toxic to human beings e.g., snake venom. The immortalization of PBMCs is induced by contact between activated cells and EBV particles. Subsequently, the memory B cells differentiate into lymphoblastoid Cell Lines which are characterized by high proliferation and antibody secretion. We often use two strategies in the immortalized PBMCs screening, which are similar to the strategies used in Antibody Affinity Maturation by Phage Display Antibody Library Screening: biopanning and solid-antigen sorting strategy.
