Conventional immunization approaches often require a large amount of purified, nonnative protein or the synthesis of several small peptide fragments of the antigen of interest. Such approaches sometimes yield monoclonal antibodies (mAbs) that are specific for improperly folded forms of the antigen of interest and have only weakly reactive with right ones. This is the common case that is extremely hard to develop high neutralizing titer and high-affinity monoclonal antibodies against the whole cells, such as an inactivated virus, by injecting the animals (hosts) with a part of antigen.

Alpha Lifetech Inc. has designed two strategies: immunization by viral antigens and I immunization by overexpress cell lines to produce high-affinity antibody.

 

Viral Antigens Immunization

 

When viral antigens as immunogens, we have set up a series of standards including GMP, GLP, ICH regulations and many strict protocols to separate and rank those antigens into different quality levels. For example, we separated one of our viral products into three levels including empty particles, incomplete particles and density particles by gradient centrifugation, which is very useful to help us identify high-immunogenicity antigens and acquire high-titer monoclonal antibodies.

 

Overexpress Cell Lines Immunization

 

We have a proprietary platform to transfect mammalian cells with extra genes that overexpress membrane proteins of interest in cell surface, where those overexpressed membrane proteins serve as "immunogens" to stimulate the host and "baits" to isolate antibodies. In this case, the antigen should remain in a pure, homogenous and stable conformational form as the native membrane protein structure. Furthermore, rich expression of target proteins in the cell surface can reduce the steric hindrance of other proteins and induce a good immune response.
