Phage Display Library –Fc-fused Single Chain Fv Construction Service
The Fc fragment brings a variety of benefits to its antibodies. Fc-fused antibodies can be simply and efficiently purified with protein A or protein G, accurately quantified, and captured in receptor-ligand affinity measurement. Fc-fused antibodies significantly improve their stability in body, because Fc fragment has the ability to reduce the effect of proteolytic degradation and keeps a long half-life period in cells. It means that Fc can increase the plasma half-life of Fc fusion proteins due to interaction with the neonatal Fc receptor (FcRn) and their larger size. While single chain Fv (scFv) offers multiple advantages over monoclonal antibodies as small molecule carriers to tumors. scFv can provide greater tumor penetration due to their small size, low kidney uptake, rapid blood clearance, and a lesser negative response by human immune system. We can provide you high-affinity Fc-fused ScFvs by using our powerful phage display technologies, including construction and screening of single chain Fv library, transient expression in mammalian cells and/or hybridoma production.
You can start your project in any step listed as bellow:
-- Generation of scFvs by synthetic DNA– This is the fastest route to construct a scFv. Oligonucleotides are first assembled in-vitro and then cloned into our in-house expression vectors. The scFv will be expressed on the surface of phage, which would be strongly recommended, since this step we can select the high-affinity antibody for you. If you wish, you can also ask us to express the fusions of interest in E. coli or a mammalian cell line for you.
-- scFv Produced from hybridoma and lymphocyte cell – The customer provides the monoclonal hybridoma cell line and this would be profect way to produce antigen-special Fc-fused antibodies. The mRNA from the monoclonal will be cloned to create a cDNA vector from which the variable heavy (VH) and light (VL) chains are then subcloned into an expression vector after the panning of VH and VL phage library.
-- scFv Manufactured from naïve sdFv library – The gene of the target antibody with high-affinity is selected from the sdFv library and cloned into our proprietary phagemid vector. We then express transiently express the fusions of interest in E. coli or a mammalian cell line for you.
