Affinity magnetic particles (AMPs) are a type of magnetic nanoparticle that is used in various applications, including biomedical research, diagnostics, and drug discovery. These particles are designed to selectively bind to specific target molecules, such as proteins or nucleic acids, and can be used for separation, purification, and detection purposes.

AMPs are typically composed of a magnetic core, such as iron oxide, coated with a layer of functionalized polymer or silica. Biomedical Applications of Affinity Magnetic Particles

 

The biomedical applications of affinity magnetic nanoparticles include in vivo applications and in vitro applications. For in vitro applications, it is mainly used for diagnostic separation, selection, and magnetorelaxometry, while for in vivo applications, it could be further separated into therapeutic (hyperthermia and drug-targeting) and diagnostic applications (nuclear magnetic resonance [NMR] imaging).

1.In vivo applications

--Therapeutic applications: Hyperthermia, the transfer of magnetic energy to the particles in the form of heat, which can be used in vivo to increase the temperature of tumor tissues to destroy the pathological cells by hyperthermia.

--Drug delivery: The particles are functionalized with specific ligands that can bind to receptors on the surface of target cells. This allows for more precise and effective delivery of therapeutic agents while minimizing off-target effects.

--Diagnostic applications: NMR imaging. Giving patients magnetic drugs can enhance the image contrast between the normal and diseased tissue and/or (2) indicate the status of organ functions or blood flow.

2.In vitro applications

--Diagnostic applications: separation and selection, magnetorelaxometry, magnetic resonance imaging and bioseparation.

--Catalysis applications: catalysts supported by MNPs have been extensively used to improve the limitations of heterogeneous catalysis.

 

Production of Affinity Magnetic Particles

 

The production of affinity magnetic particles requires careful consideration of factors such as particle size, magnetic properties, and functionalization. Ongoing research and development are expected to further enhance the performance and versatility of AMPs in the future.

 --Co-precipitation method: iron salts are precipitated in the presence of a functionalized polymer or silica. The size and magnetic properties of the particles can be controlled by adjusting the reaction conditions, such as the concentration of the reagents and the reaction time. The co-precipitation method could produce AMPs with high binding capacity and specificity.

  --Emulsion method: a water-in-oil emulsion is formed by functionalized particles dispersed in the aqueous phase. The emulsion is then mixed with a cross-linking agent to form a solid gel, which is subsequently broken down to release the particles. The emulsion method could produce AMPs with high stability and biocompatibility.

  --Microfluidic reactors: microfluidic reactors offer several advantages over traditional batch reactors, including improved control over reaction conditions and reduced waste. Microfluidic reactors could produce highly uniform AMPs with improved binding capacity.

Affinity magnetic particles (AMPs) can be used for affinity purification and separation of antibody, antigen, proteins and DNA/RNA, etc. AMPs have high selectivity and capacity, which make them ideal tools to separate of various biomolecules. Alpha Lifetech Inc. provides uniform and monodisperse affinity magnetic particles, available in nanometer (50 nm, 100 nm), submicron (150 nm, 200 nm) and micron (0.5-12 μm).

Alpha Lifetech Inc. can combine a variety of bioaffinity ligands with magnetic beads to meet customer needs. These ligands include, but are not limited to, antibodies, biotin, GST, streptavidin and lectins. Nano and submicron functional magnetic beads are paramagnetic, and the monolayer bovine serum albumin (BSA) is covalently coupled to the surface of the magnetic material, which makes the surface properties of the magnetic beads better, the stability enhanced, and the non-specific binding reduced. Micron-sized functional magnetic beads are paramagnetic and consist of cross-linked polystyrene and uniformly distributed magnetic materials. Abundant affinity ligands are covalently introduced to the surface of the magnetic beads, so that the affinity magnetic beads have more biological binding sites, the amount of magnetic beads used is small, and the non-specific binding rate is low.
